Neuro D (also designated BETA 2) is a basic helix-loop-helix (bHLH) transcription factor functioning in neurogenic differentiation. Neuro D is expressed in pancreatic islet cells and transiently in a subset of neurons in the central and peripheral nervous systems at the time of their terminal differentiation into mature neurons. Moreover, ectopic expression of Neuro D in Xenopus embryos induces premature differentiation of neuronal precursors, and Neuro D can convert presumptive epidermal cells into neurons. BETA 3 is a novel helix-loop-helix protein that can function as a negative regulator of both Neuro D and Myo D-responsive genes. BETA 3 exhibits the highest degree of sequence relatedness with Neuro D and is most highly expressed in lung, kidney and brain cells.

The pASG-IBA43 vector is designed for expression of recombinant proteins with an N-terminal His-tag and a C-terminal Strep-tagII in E. coli. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells, F1 and ColE1 origin for a high plasmid copy number, and an inducible tetracycline promoter/operator for the regulated expression of proteins. Expression of the recombinant protein is induced by addition of anhydrotetracycline, which is a derivative of tetracycline that does not exhibit antibiotic activity. Please note that cloning into pASG-IBA Acceptor Vectors compulsorily requires the restriction enzyme Esp3I since no other MCS for the integration of a gene of interest is available. In addition to the direct cloning of the gene of interest into pASG-IBA vectors with Esp3I, another option via a so-called Entry Vector is possible.

Vector Biolabs AAV-CreERT2 (AAV serotype 1) AAV (AAV1-CreERT2)
A mutated form of estrogen ligand-binding domain (ERT2) that binds to synthetic antagonists (such as tamoxifen or its derivative 4-hydroxy-tamoxifen) but not to circulating estrogens was fused to the Cre recombinase (Cre) to create the CreERT2, which requires the presence of tamoxifen for Cre recombinase activity. Therefore it makes possible to induce the recombination in cells with tamoxifen.This AAV-1 virus expresses CreERT2 under a CMV promoter.

pBR322 DNA is a commonly used plasmid cloning vector in E. coli and is a double-stranded circle 4,361* base pairs in length. pBR322 contains the genes for resistance to ampicillin and tetracycline, and can be amplified with chloramphenicol. The molecular weight is 2.83 x 10^6 daltons.
